Abstract : | [Abstract] The use of ruthenium red as a stain for electron microscopy of external cell surfaces was tested using cultured rat osteosarcoma cells, ROS/17. In situ staining of cell cultures with osmium tetroxide and 0.01, 0.05 or 0.1% ruthenium red, which had been left to oxidize for 3 hours prior to use, resulted in well defined cell boundaries. The stain penetrated consistently into the closely opposed intercellular junctional areas. If section staining by uranyl acetate-lead citrate is also done, intracellular components can be identified. This clear definition of extra-and intracellular surfaces will facilitate quantitation of the cell ultrastructure. |
