Abstract : | [Abstract] The effect of mechanical stretching on an alkaline phosphatase (ALPase) activity was studied by bone cell culture. Primary cultures of bone cells, isolated from a 2-day-old rat calvaria, were subcultured in dishes with a flexible plastic bottom. The attached cells were stretched by means of the method described by Hasegawa et al., 1985. The ALPase activity was increased by an intermittent stretching of confluent cells. The results of SDS-PAGE showed that the increased ALPase activity raised in the 110K band. This stretch-responsive ALPase was bound to hyaluronic acid. Hyaluronic acid synthesis increased in the stretched cells. These results indicate that the mechanical stretch-sensitive ALPase is a hyaluronic acid binding, 110K ALPase. |
