| Abstract : | [Abstract] Testosterone (T) metabolism and the effects of sex hormones on the growth were investigated in the squamous carcinoma (RSS) cell strain of the rat submandibular gland (SMG) induced by 7,12-dimethylbenz[a]anthracene (DMBA). A large amount of dihydrotestosterone (DHT) was produced from the substrate T in the RSS cell. The higher activity of 5α-reductase was revealed in the RSS than normal cells. The DHT formations from the substrate were 8.3pmol/l×105 cells for normal cells and 17.5pmol for the RSS cells, when 3H-T was incubated at the concentration of 7.2μM for 20 min. The DHT formation in sarcoma (RFI) cell was similar to normal. Δ4-Androstenedione (Δ4-Dione) production, however, was lower in the RSS than normal. The growth of RSS cells was accelerated at the T concentration of 10-7μM and the growth showed a plateau at 10-4μM, showing 1.8-fold on nontreated cells. The growth of the cells was increased 1.5-fold by 10-4μM of estradiol-17β (E2) as compared with nontreated cells. The cell growth was not influenced by progesterone (P4). These results mean that T metabolism exists in the carcinoma possessing high 5α-reductase activities in the cells and that the growth of the tumor cells is controlled by androgen. |
