Abstract : [Abstract] Interleukin-2 (IL-2) with anti-CD3 antibody can induce numerous lymphokine activated killer (CD3LAK) cells. However, CD3LAK cells exhibit less cytotoxicity than LAK cells treated with IL-2 alone. We estimated the enhancement of cytotoxicity by PHA or PWM against peripheral blood mononuclear cells (PBMC)-CD3LAK cells derived from peripheral blood mononuclear cells from a clinical healthy volunteer as well as PBMC-CD3LAK and tumour infiltrated lymphocytes (TIL)-CD3LAK cells obtained from two patients with tongue cancer (stage II). PHA or PWM added for assay period (4 hours) of cytotoxicity in microplate. Cytotoxicity was estimated with a 51Cr release assay. We found that NK activity of the PBMC-CD3LAK cells derived from a clinical healthy volunteer was significantly enhanced by PHA or PWM. LAK activity of the above cells or PBMC-CD3LAK cells derived from case 2 was enhanced significantly only by PWM. The NK activity of TIL-CD3LAK cells derived from case 1 was enhanced by PHA or PWM, and the LAK activity of these cells was enhanced significantly by PWM. The LAK activity of TIL-CD3LAK cells derived from case 2 was enhanced by PWM. As for cytotoxicity against the autologous tumour, PBMC-CD3LAK cells derived from case 2 were also slightly enhanced by PHA or PWM, but TIL-CD3LAK cells enhanced significantly more than the PBMC-CD3LAK cells in this study. These results suggest differences between PBMC-CD3LAK and TIL-CD3LAK cells in the repertoire and in the sugar chain expressed on the tumour cell surface.
