Abstract : | The wall-less procaryote Mycoplasma fermentans (Mf) is currently being examined as an agent potentially associated with human disease. To delineate possible roles of Mf or Mf-derived antigen (TXLP) on generation of the temporomandibular disorders (TMD), we examined the ability of TXLP to induce cytokine as well as matrix degrading enzyme (MMP) production on various cells, which composed of articular tissues. TXLP significantly induced TNF-α and IL-1β production on human blood cells, but no stimulatory effect was observed on human chondrocytes. MMP-3 mRNA expression was also induced on rabbit synoviocyte by TXLP stimulation. Furthermore, receptor for Mf lipoprotein (Toll-like receptor2: TLR2) was highly expressed on human blood cells. As the most interesting finding, we found existence of Mf-derived DNA in 44 out of 120 (36.7%) TMJ synovial fluid (SF) samples. These results together with our previous data suggest that Mf would have potential to induce arthritis in the TMJ, which may proceed to TMD. |
