Abstract : | [Abstract] In order to elucidate biochemical markers to detect early cartilage degradation in the temporomandibular joint (TMJ), we examined matrix metalloproteinase (MMP) activities in the synovial fluid (SF). SF was obtained from 13 TMJs from osteoarthritis (OA), 22 with internal derangement (ID), and 13 from asymptomatic normal volunteers (NR). MMP activities were detected by gelatin zymography and Western blotting using specific antibodies to various MMPs. We identified MMP-9, -2, -3, and -1 activities in the SF using gelatin zymography and Western blot analysis. Among these, the 83kDa, 66kDa, 41kDa, and 45kDa bands that were considered to be active-forms of the aforementioned MMPs were detected. The intensity of the 83kDa (active-MMP-9) was significantly higher (p<0.05: Mann Whitney U-test) in OA and ID patients when compared with NR patients. Furthermore, the frequency of MMP-3 detected in the OA-SF was significantly different (p<0.05: Chi square test) from that of the NR group. These findings suggest that an increase in the active forms of MMP-9 and the appearance of MMP-3 in SF may be potential catabolic markers for cartilage degradation in the TMJ. |
