Abstract : | [Abstract] Orexin-A (hypocretin-1) and orexin-B (hypocretin-2) neuropeptides are located in the lateral hypothalamus and dorsomedial hypothalamic nucleus, and these neurons project to a wide range of locations in the rat, including the cerebral cortex, medial groups of the thalamus, circumventricular organs, the hippocampus and amygdala within the limbic system, the locus ceruleus, and the raphe nucleus. Orexin neurons have been implicated in diverse functions, including energy homeostasis, neuroendocrine systems, sleep-state cycle regulation, and feeding behavior. These actions of orexins are mediated via two different G-protein-coupled receptors (orexin 1-receptor, orexin 2-receptor). The orexin 1-receptor has been thought to be selective for orexin-A peptide, whereas the orexin 2-receptor has been bound to orexin-A and orexin-B peptides with similar affinities. In this study, double immunoreactive orexin-A and orexin-B immunoreactive fibers and varicose structures were found in the same fibers. And orexin 1-receptor immunoreactive cells were found within neurons in the trigeminal motor nucleus, and orexin-A immunoreactive fibers were found sparsely with button-like structures in the trigeminal motor nucleus. The distribution of orexin fibers and receptors in the trigeminal motor nucleus suggests that orexin neuropeptides not only play a role in the synaptic control of orexin motoneurons, but that they also play other physiological roles related to food-taking behavior. |
