Abstract : [Abstract] An electron microscopic observation on the secretion granules of the submandibular glands of adult female rats revealed a mechanism of granular release similar to macroapocrine secretion. The merocrine, reverse pinocytosis or exocytosis secretion of release so far recognized and accepted were not encountered in the present study. As the secretory granules make contact with the plasma membrane on the freecell surface, the limiting membrane surrounding the granules was fused by the plasma membrane. The process up to this stage may be explained as merocrine secretion. As the protrusion into the glandular lumen advances, a limiting membrane is found on that part of the secretory granule that faces the lumen, and this limiting membrane is fused with the plasma membrane. On the part facing the cell's basal surface, on the other hand, the limiting membrane disappears and cannot be demonstrated. Finally, the secretory granule protruding into the lumen is surrounded by the limiting membrane. Based on these findings, it is assumed that the limiting membrane is at one time fused to the plasma membrane and then formed anew by the plasma membrane in the new concept.
