Abstract : [Abstract] The present study demonstrated that the extract from Ulex europeus seeds contains a substance or substances that inhibits the human lymphocyte DNA, RNA and protein synthesis. This inhibition is proportional to the metabolic rate of the lymphocytes and the length of exposure to the Ulex seed extract. Mitotic stimulation as varied as PHA, Con A, mixed lymphocyte reaction and rapid growth of lymphoblasts are equally susceptible to this inhibition. The inhibitory factor was not related to anti-H activity. Lymphocyte- and platelet-agglutinin were not observed when tested by usual procedures. Attempts to characterize the inhibitory substance (s) showed that it was not cytisine. Physichochemical studies of the extract showed that the inhibitory factor was heat-stable. The results suggest that the substance was a glycoprotein. It is suggested that there is a receptor in the lymphocyte membrane which may be activated by Ulex europeus seed extract. Subsequently, the activated membrane receptor prevents the metabolic events which mitogens normally produce. In addition, the same activated receptor may turn off a cell already committed into a metabolic stage and bring it back to its resting stage. This inhibition appeared to be reversible when the extract was removed. It is concluded that the Ulex europeus inhibitor (UEI) produces its effects only as it is weakly bound to the lymphocyte receptors. These results suggest that the Ulex europeus seed extract contains an inhibitor or inhibitors which are counterparts of the well-known stimulatory substances present in other plant extracts.
