Abstract : Since Stre.mutans are grown in a brain heart infusion (BHI) and tryptic soy broth (TSB), these media were used for cultivation of the bacteria. In cultivating Stre.mutans in BHI and TSB, the organisms in the media lose their activity a few days after the beginning of the cultivation. Hence, death of the cells after a short period always troubles researchers treating the organisms because they want the bacteria to survive for a long period. For this purpose, the authors prepared a medium containing sodium alginate and cultivated the organisms in the media. Two media were prepared, one of them was BHI containing sodium alginate and other, TSB also containing sodium alginate. Experimental procedures follow. Sodium alginate (Kanto Chemical Co.) of 0.005, 0.1, 0.5, 1.0 and 3.0% was added to BHI (brain heart 0.6%, thiotone peptone 0.6%, sodium chloride 0.5%, dextrose 0.3%, gelysate peptone 1.5% and disodium phosphate 0.2%, BBL) and to TSB (tryptone 1.7%, soytone 0.3%, dextrose 0.25%, sodium chloride 0.5%, dipotassium phosphate 0.25%, Difco).
