Abstract : When Stre. mutans was cultivated in a medium containing sugar, the organisms died out almost in a week. For keeping the survival period of the acidogenc bacteria as oral streptococci and lactobacilli, some treatment of the medium is necessary. One is to add a reagent like sodium alginate, calcium chlorides and CaCO3 to the medium. Therefore, in this experiment the authors examined the relation between a concentration of calcium, phosphate in the medium, a live period of the bacteria cells and the range of pH during the incubation period. From the results it was found that when a certain concentration of the reagent was put into the medium, the cultivating cells survived for some period. [Experimental Materials and Methods] 1. Bacteria: Stre. mutans E49 was used in this experiment. 2. Medium: Tryptic soy broth (Difco) was used as the base medium of this experiment and a different concentration of calcium phosphate (CaPO4 2H2O) (0.01, 0.05, 0.1, 0.5, 1.0% and the control group not containing the reagent) was included in the medium. Next, 30ml of the medium was poured into the test tubes 30mm in diameter.
