Abstract : [Abstract] The existence of 4S androgen binding protein in the cytosol of human submandibular gland (SMG) was demonstrated. Tissue samples were obtained from patients with cancer of the tongue, oropharynx cancer and sialoadenitis submandibularis. Samples were incubated for 20 hours at 4℃, and dextran-coated charcoal was used to separate bound steroids from free steroids. The presence of 3H-dihydrotestosterone (3H-DHT) binding protein was demonstrated in the cytosol fraction of human SMG by 5-20% linear sucrose density gradient. This complex had an approximate sedimentation coefficient of 4S. DHT was a more potent inhibitor of 3H-DHT binding than testosterone (T) and methyltrienolone (R 1881) in the human SMG cytosol. However, the ability of DHT to compete with 3H-R 1881 binding was not so potent as R 1881 and the inhibitory effect of DHT on 3H-R 1881 was similar to that of T.
