Abstract : | [Introduction] Recently Kodama et al., following the subculturing of cells from a newborn mouse calvaria, produced a cell line called MC3T3-E which showed a strong alkaline phosphatase activity and accordingly established a clonal osteoblastic cell line, MC3T3-El, which have retained the high phosphatase activity as its primary indication. After a one- or 2-week culture, numerous collagen fibrils intermingling in the intercellular space could be identified, and a calcification process very similar to that under in vivo conditions also was observed after a 3-week culture. Several biochemical investigations hitherto on the characteristics of these clonal cells responding to hormones enabled us to understand the role of osteoblasts in vivo. The detailed information about the morphological criteria, however, was very limited, as compared to those conducted by biochemical approaches. Under in vivo conditions the osteoblasts were shown to phagocytose the extracellular collagen fibrils. Only a few reports which were conducted under in vitro conditions indicated that a large number of collagen or collagen precursor containing vacuoles were observed in the osteoblasts. |
