Abstract : [Precursors of Osteoclasts] The osteoclast is the unique multinucleated cell responsible for normal and pathological bone resorption. Although it is known to be derived from hematopoietic cells, its lineage is controversible. Transplantation experiments with bone marrow in human and rodens have shown that the osteoclast precursors are present in bone marrow cells. Marks and coworkers and others have also shown that peripheral blood or spleen cells in rodents contain osteoclast precursors and transplantation of these cells into affected rodents can cure osteopetrosis. However, the specific cell lineage from which the osteoclast is derived is in dispute. Several lines of evidence suggest that the osteoclast is derived from the macrophage lineage. Burger et al., using a mouse marrow culture system, have showed that the precursor cells expressed macrophage antigens. Baron et al. and Roodman et al. showed that the precursors contain nonspecific esterase, which is present in monocytes. Further, Roodman et al. suggested that the precursor is an immature monocyte by morphologic studies.
