Abstract : | [Abstract] The growth of normal fibroblasts is in need of an adhesive substratum on which to spread. An attachment activity was measured because it has been shown to be required for growth and spreading of human periodontal ligament fibroblasts (HPLF). The degree of spreading was controlled by coating the conditioned medium in hydrophobic culture plates. This medium was derived from HPLF exposed to varying magnitudes of mechanical stress, comprised of 3 cycles per minute of 0, 6, 18 and 24% elongation. HPLF immediately transformed and spread in the culture plate by coating the conditioned medium in the stress culture compared with the blank. Surprisingly, HPLF were rapidly transformed and spread by the conditioned medium of 24% stress more than that of 6% stress. The present data suggest that the requirement for spreading may in part by mediated be mechanical stress. |
