Abstract : [Introduction] The periodontal ligament is derived from the dental follicle which consists of undifferentiated cells. The restricted transit cells differentiated from periodontal stem cells as shown in Fig. 1 probably predominate in the periodontal ligament and could regulate the expression of the phenotypes for bone and cementum by osteoblasts and cementoblasts at the alveolar bone and cementum surface, respectively. Melcher et al. and Arzate et al. have demonstrated that alveolar bone and cementum may be the same or similar tissues. With increasing pace among the clinical and basic researchers who studied the fundamental character of periodontal ligament, there has been the development of the cell culture system of fibroblasts derived from human periodontal ligament. We have cultured cells (HPLF-Y and HPLF) from explants of the human deciduous and permanent periodontal ligaments and characterized the osteoblastic phenotypes of HPLF. In this review, we focused on (1) the fundamental characters of HPLF in vitro, (2) the effect of vitamin D3 on the ALPase expression of fibroblasts derived from the three different developmental stages of bovine dental follicle, and (3) the comparison of HPLF-Y and HPLF incubated with vitamin D3, dexamethasone, EGF and TGF-β1.
