Abstract : [Abstract] The abnormality of the tumor suppressor gene, p53 and the RB genes, was studied in human cancer cell lines derived from cancer of oral cavity and associated cancer, using the methods of single strand conformation polymorphism (SSCP) analysis of RNA and polymerase chain reaction (PCR)-quantitation. We found the structural alteration of the p53 gene and RB gene in KB (squamous cell carcinoma of oral cavity) cells and HMV-1 (malignant melanoma) cells, and the absence of RB gene expression in HMV-1 cells from the results of PCR-quantitation. These results suggest that the structural alteration and the abnormal expression of p53 and RB genes could be involved in the development of cancers of oral cavity. The importance of the validation to analyzing the tumor supressor genes in clinical samples of human oral cancers are also indicated.
