Abstract : [Abstract] A squamous cell carcinoma cell line (RSS18) derived from a 7, 12-dimethylbenz[a]anthracene-induced submandibular gland of a femal rat was tested for production of ECM-degradative proteinases. Zymographic analysis showed that gelatinolytic activities associated with 85-and 25-kDa proteins were secreted in a serum-free medium. Enzyme-inhibition assays were performed to determine the proteinase type. The gelatinolytic activities of the 85-and 25-kDa proteins were completely inhibited by diisopropyl fluorophosphate, suggesting that they belong to the serine proteinase type. When the conditioned medium was treated with enteropeptidase at 37℃ for 10min before zymographic analysis, the gelatinolytic activity of the 25-kDa protein significantly increased. Moreover, reverse transcription-polymerase chain reaction analysis revealed that trypsinogen mRNA was expressed in the cells. These results suggested that 85- and 25-kDa proteins were similar to plasmin and trypsin proteinases, respectively.
