Abstract : [Abstract] The distal portion of the prepubertal rat submandibular gland consists of terminal tubules. In this portion, three types of cells were observed : acinar cells (A-cells), proacinar cells (PA-cells), and terminal tubule cells (TT-cells). An intercalated duct and a striated duct appeared continuously with the terminal tubules. Electron microscopy showed the absence of PA-cells from 14 days after birth. Paraffin sections stained with the terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling (TUNEL) method showed negative reactions in all cells until 14 days after birth. The TUNEL method showed positive reactions in a small number of cells in the TT-cells. The number of positive cells increased until 28 days after birth. Electron microscopy revealed nuclear chromatin aggregation, suggesting apoptosis in TT-cells 21 days or more after birth. Phagocytosis of the apoptotic body by intercalated duct cells was observed. These results suggest the close involvement of apoptosis in the regulation of cells constituting the submandibular gland in the postnatal growth of the rat submandibular gland.
