Abstract : [Abstract] The purpose of this study was to investigate the neuropeptide contents of neurons labeled with Di-I from an intact junctional epithelium (JE). Eight Sprague-Dawley male young adult rats were used. After general anesthesia, Di-I was applied to the buccal sulcus of the upper first molars; and after seven days the rats were sacrificed. The excised trigeminal ganglia (TG) were cut in serial frozen sections along the horizontal plane. All sections were then reacted using immunocytochemistry for calcitonin gene-related peptide (CGRP) and substance P (SP). All Di-I labeled neurons (red) were in the maxillary region of the TG, and 77.2% of them had labeled CGRP and 31.2% had SP. The diameter of the neurons was 37.2+-11.42μm of only Di-I labeling, 32.1+-9.65μm of CGRP-Di-I double labeling, and 27.4+-6.98μm of SP-Di-I double labeling. Hence we conclude that the neurons which innervate gingival tissue are predominantly peptidergic and can take up and transport exogenous substances such as Di-I to the cell body.
