Abstract : [Abstract] Technetium-99m-hydroxymethylene diphosphonate (99mTc-HMDP) has been more frequently used than Technetium-99m-methylene diphosphonate (99mTc-MDP), because 99mTc-HMDP has been reported to be more accumulated in the bone. However, no crucial factor involving such difference has been identified. Thus the present experiments were carried out to determine the cause of the difference in accumulation. Compared with 99mTc-MDP, 99mTc-HMDP was markedly adsorbed to hydroxyapatite both in vivo and in vitro. When hydroxyapatite-adsorbed 99mTc-MDP and 99mTc-HMDP were rinsed with diphosphate, more than a half of 99mTc-HMDP remained in hydroxyapatite, while more than a half of the 99mTc-MDP was washed away from the hydroxyapatite. However, the stability of 99mTc-HMDP in distilled water or blood was poorer than that of 99mTc-MDP.
