Abstract : [ABSTRACT] The lipopolysaccharide (LPS) of Veillonella parvula has the high potential effects on the osteoclast formation. The effects of LPS receptor blocking on the bone-resorbing ability of osteoclasts induced by V.parvula LPS were examined to clarify the expression of CD14 on osteoclast. In addition, we investigated whether cytokines can relate to bone resorption of osteoclast induced by V.parvula LPS. Mouse bone marrow cells were cocultured with MC3T3-G2/PA6 cells on ivory in the presence of V.parvula LPS. The addition of anti-CD14 antibody decreased the resorption area after 7 days culture. The addition of bone resorptive cytokines (IL-1α, TNFα and IL-6) also weakened bone-resorbing ability of the osteoclasts. These results suggested that CD14 on osteoclasts may act as a receptor for LPS, and that LPS may enhance the cytokine production of osteoclasts.
