Abstract : [Abstract] The specific aims of this investigation were to detect type II procollagen C-peptide (pColl-H-C; chondrecalcin) from the synovial fluid (SF) of tempromandibular disorders (TMDs) and analyze its correlation with inflammatory cytokines (IL-1β, TNFα) or matrix degrading enzymes (MMPs). SF was obtained from 15 TMJs (7 osteoarthritis (OA), 8 internal derangement (ID)) in 15 patients and 8 normal asymptomatic TMJs of 8 volunteers (NR). Measurement of pColl-II-C, IL-1β and TNFα in the SF were performed by enzyme immunoassay (EIA). MMP activities were detected on gelatin enzymogram and type of MMPs was identified by Western blot analyses. The concentrations of pColl-II-C in SF were 1.O4, 0.39 and 2.59ng/ml in NR, ID and OA, respectively. The level of pColl-II-C in OA was significantly higher (p=0.029) than that of NR and ID. There was no significant correlation between the level of pColl-II-C and that of MMP2, MMP9, IL-1β or TNFα. These results suggest that TMJ OA is associated with increased synthesis of type II collagen accompanied with cartilage repair.
