Abstract : [Abstract] Cementoblasts have been believed to differentiate from dental follicle cells which contained progenitors of periodontal cell lineage. To elucidate the molecular mechanisms of cementogenesis, we have examined the characterization of dental follicle cells if they contained cementoblasts progenitors. The bovine tooth germ at root forming stage was digested with bacterial collagenase, and the cells released from the tissue were maintained as bovine dental follicle cells (BDFC). We analyzed the expression of extracellular matrices involved in cementogenesis including bone sialoprotein (BSP), osteocalcin (OC), osteopontin (OP) and type I collagen (COLI) using RT-PCR and measured alkaline phosphatase activity. We also examined the expression of Cbfa1/Pebp2αA, a crucial transcription factor for ossification. Results showed that after 5 days incubation confluent BDFC expressed BSP, OC, OP and COLI, and showed alkaline phosphatase activity. Interestingly, the expression of Cbfa1/Pebp2αA was detected in BDFC. These data indicate that BDFC contain cementoblast progenitors which express extracellular matrix components related to cementogenesis, and also transcription factors related to membranous ossification.
