Abstract : [Abstract] It has been clarified that topoisomerase type IIα (topo IIα) plays an important role in DNA metabolism including transcription, replication and proliferation. Topo IIα immunostaining is the newest proliferative marker and can utilize paraffin embedded sections. We conducted our research to discover whether the immunohistochemical localization of topo IIα was specific in cell proliferation layers in seven cases of oral squamous epithelia at the buccal mucosa, using as an index the distribution of DNA content shown with an Anchored Cell Analysis Sorting 570 (ACAS 570) laser cytometer with confocal laser scanning microscopy. Immunolocalization of topo IIα was identified in the nuclei of the oral squamous epithelia in the basal and parabasal cell layers in all cases. The distribution of topo IIα-positive cells was in accord with the high DNA content on the section analyzed using an ACAS570 laser cytometer. Although there was a close correlation between the distribution of topo IIα and the high DNA content in the serial sections, topo IIα immunostaining was useful as a proliferation marker in the stratified squamous epithelia of the oral mucosa. Topo IIα immunostaining may be expected to have some significance as a proliferative marker for oral squamous lesions.
