Abstract : [Abstract] SPARC (secreted protein, acidic and rich in cysteine) is thought to be associated with angiogenesis and tumor growth. In the present study, we determined SPARC expression in cells from several carcinomas of the head and neck. Reverse Transcription-Polymerase Chain Reaction analysis showed that SPARC was expressed in 4 cell lines derived from tongue carcinomas but not in 3 cell lines from carcinomas of the hypopharynx, oropharynx, and oral floor. In the 16 clinical specimens containing tongue carcinoma cells, we detected SPARC in tumor cells from 9 cases and in the adjacent endothelial cells (EC) from 12 cases. Especially, SPARC was strongly expressed in EC adjacent to tumors classified into Stage III-IV. Furthermore, we found that VEGF induced SPARC expression in EC. Our findings suggested that SPARC contributes progression of tongue carcinoma.
