Abstract : | Pebp2αA/cbfa1 is a member of the runt family of transcription factors. It has been indicated that pebp2αA/cbfa1 plays an essential role in osteoblast differentiation and skeletal patterning. Pebp2αA/cbfa1-deficient mice completely lacked bone formation in both intramembranous and endochondral ossification. We have raised monoclonal antibody against recombinant murine pebp2αA protein. The specificity of this antibody was characterized by its reaction in Western blot analysis and electrophoretic mobility shift assay (EMSA). Using immunohistochemical methods we have also investigated the localization of pebp2αA/cbfa1 in the bone and teeth of rats of different developmental stages. Pebp2αA/cbfa1 protein was localized in the nucleus of osteoblasts, cells in endosteal tissues, hypertrophic and proliferative chondrocytes of rat mandibular condyle. Dental follicle cells also showed a positive reaction to the pebp2αA/cbfa1 antibody. At day 18 and 20, the protein was located in the nuclei of mature ameloblasts and cementoblasts. No immunostaining was detected in the osteoclasts or osteocytes in alveolar bone. Our findings indicate an important role for pebp2αA/cbfa1 in skeletal development, tooth formation and osteogenic differentiation of mandibular condyle. |
