Abstract : Concanavalin A(Con A)induced hepatitis is an experimental model of T cell dependent autoimmune hepatitis. We examined the role of LFA-1 in Con A-induced hepatic damage using LFA-1-/-mice. Massive hepatic necrosis-like regions were observed in all LFA-1+/+ mice. In contrast, LFA-1-/- mice were resistant to Con A-induced hepatitis and none of LFA-1-/- mice showed hepatic necrosis-like regions. Whereas activated liver T cells remained in the liver in LFA-1+/+ mice, activated T cells were rapidly cleared from the liver of LFA-1-/- mice. Liver T cells from LFA-1-/- mice showed impaired adhesion to murine hepatocytes in vitro. Adoptive transfer of liver T cells from LFA-1+/+ mice, but not from LFA-1-/- mice, sensitized LFA-1-/- mice to Con A-induced hepatitis. These data indicated that LFA-1 mediated cell to cell interaction between hepatocytes and T cells are crucial for Con A-induced massive hepatitis.
