Abstract : Cementum-derived attachment protein (CAP) has been reported as a collagenous attachment protein existed in cementum. Recently, we have shown that the CAP was a putative cementoblasts marker in developing tooth germ, and it may contribute cementogenesis through the promotion of cell attachment to dental follicle cells. To identify the molecular structure of CAP, it was extracted from bovine tooth germ, and separated by diethylamino ethyl (DEAE)-cellulose chromatography and Q-Sepharose fast flow column, and purified by Mono Q high performance liquid chromatography (HPLC). The purified preparation contained single band migrating with molecular mass of 65kDa. The 65kDa protein was positive for anti-CAP monoclonal antibody, and it was susceptible to bacterial collagenase. These data indicated that CAP was able to purify from bovine developing tooth germ.
