Abstract : The effects of minor fimbriae of Porphyromonas gingivalis which consisted of 67-kDa fimbrillin on IL-1β production in murine peritoneal macrophages and induction of osteoclast differentiation were investigated. Murine peritoneal macrophages were stimulated with 67-kDa minor or 41-kDa major fimbriae, and the levels of IL-1β production were determined by ELISA. To estimate osteoclast differentiation, mouse osteoclast precursors were placed on dentine slices, and cultured with or without fimbriae for 7 days. Both minor and major fimbriae explicitly induced IL-1β production in murine macrophages. Furthermore, pit formations on the dentin slices were significantly enlarged when the osteoclast precursors were incubated with P.gingivalis fimbriae. Pretreatment of murine macrophages with anti-TLR2 and/or anti-TLR4 antibodies inhibited IL-1β activity stimulated with both fimbria. These results suggest that minor fimbriae of P.gingivalis may provoke host inflammatory response as well as major fimbriae and be involved in the development of periodontal disease.
