Abstract : A 16S rRNA-based PCR detection method was used to determine the prevalence of Porphyromonas gingivalis and Porphyromonas gulae in subgingival specimens from dogs and humans. P.gingivalis only detected the specimens from humans. However, P.gulae detected the specimens from dogs and humans. The fimbrial protein was purified from Porphyromonas gulae ATCC 51700 by DEAE-Sepharose CL-6B column chromatography. The fimbrial protein was eluted at 0.15 M NaCl, and the molecular mass of this protein was approximately 41K as estimated by SDS-PAGE. Immunogold electron microscopy showed that the anti-41K fimbriae serum bound to the fimbria on the cell surface of P.gulae. The component of the fimbrial structure (fimbrillin) of P.gulae ATCC 51700 was the same in size and in antigenicity as that of 41K major fimbrial protein of P.gingivalis ATCC 33277. In addition, the presence of P.gulae was also found in human oral cavity.
