Abstract : [Abstract] The present study was initiated to answer the question "Does properties of dentin that was initially demineralized by microcosm biofilm influence the bacterial metabolism?" To investigate this, biofilms were formed in a high-throughput active attachment biofilm system. The aims of this study were as follows: 1. to validate a model of mature microcosm biofilms formation (8 days), which simulated bacterial metabolism on glass or dentin substrates. 2. to prove the hypothesis that the metabolism of the microcosm biofilm is affected by the characteristics of the substrate it grows on. Biofilms formed on glass or dentin surfaces were harvested at several time periods (48, 120, 144, 168, and 192 hrs) and their metabolism, based on pH of the medium and colony-forming unit (CFU) counts, was analyzed. The results showed that biofilms formed on glass and dentin differ in all properties measured. In addition, biofilm formation on dentin caused stable calcium release. The data collectively illustrate the reliance of long term microcosm biofilm model and the importance of the choice of substrate for biofilm growth when restoring the oral cavity.
