| Abstract : | 「緒言」 マウス, ラット顎下腺の組織学的所見および, 顎下腺内のホルモン様生理活性物質に性差が存在し, しかもこれらは, Androgen依存であると言われている. さらにマウス顎下腺のサイトソールに, 活性型Androgenと言われる5α-Dihydrotestosterone(DHT)と特異的に結合するタンパクが存在することも報告されている. これらの事実より, 顎下腺がAndrogen標的臓器であると言われ, 近年注目されつつある. 一方, Androgen標的臓器においては, Tを還元し, DHTを生成する5α-Rが重要な酵素とされており, それぞれの臓器内における5α-Rの性質によりその臓器のAndrogen依存性の決定および方向づけがなされている. そしてラット顎下腺にT代謝が存在し, この5α-Rについては, 雌ラット顎下腺マイクロゾームでは, Tに対するKm値は1つ存在するのに対し, 雄のそれは2つ存在し, しかも雄の一方の低い値のKm値は, 雌のKm値と同じであることが報告されている. |

| Abstract : | Abstract : The properties of 5α-reductase (5α-R) in the submandibular gland (SMG) of rats were investigated using microsomal fraction in the presence of NADPH. It is known that the formation of 5α-dihydrotestosterone and 5α-androstane-3α, 17β-diol and that two apparent Michaelis constants (Km) of this enzyme for testosterone are obtained in SMG of male rats. Rats castrated at the age of 3 or 7 weeks were sacrificed at 7 or 11 weeks of age respectively, and their SMG were investigated. Microsomal fraction of SMG from uncastrated male rats and from castrated ones, each of which was incubated with 14C-labeled testosterone. The activities of 5α-R in castrated rats were effectively decreased as compare with uncastrated rats. Two Kms obtained as 0.92×10-6M and 12.05×10-6M in the rat castrated at 7 weeks of age showed similar to the values in uncastrated male rats. Whereas only one Km obtained as 1.16×10-6M in rats castrated at 3 weeks of age was similar to that in females. The present study demonstrated that gonadoectomy of male rats suppressed the activity of 5α-R in microsomes of rat SMG and that the property of 5α-R possessing two Kms for testosterone in uncastrated male rats appeared after 3 weeks of age. |
