| Abstract : | 「緒言」 Porphyromonas gingivalis(P.gingivalis)は偏性嫌気性の黒色色素産生グラム陰性桿菌であり, 成人性歯周疾患との関わりが強く示唆されている. 本菌の表層には, 繊維状構造物で歯周組織細胞への付着性に関与していると考えられている線毛や血球凝集素の他, コラゲナーゼやトリプシン様酵素など, 直接的に組織破壊を引き起こすタンパク分解酵素, あるいは, 好中球やマクロファージによる貪食に抵抗性を示す莢膜などが存在している. また, グラム陰性菌の特徴の一つである外膜は, 菌体内に親水性物質を取り込む働きをするポーリンタンパクなどの外膜タンパクや内毒素(lipopolysaccharide : LPS)から構成されている. LPSは発熱作用や, 致死毒性を示す他, 免疫細胞を活性化したり, 歯周組織細胞からサイトカインの産生を誘導することが知られている. |

| Abstract : | Abstract : We compared the immunobiological activities of the 75K protein, fimbrial protein, and lipopolysaccharide (LPS) which were surface components from P.gingivalis. The 75K protein was mitogenic for BALB/c nu/nu, BALB/c, and LPS-responsive C3H/HeN mice spleen cells and it was essentially mitogenic for LPS-nonresponsive C3H/HeJ mouse spleen cells. The 75K portein and LPS were explicitly mitogenic at a concentration of 1 μg/ml (P<0.05) in BALB/c mouse spleen cells but there were no apparent effects on thymocytes. Furthermore, the 75K protein exhibited polyclonal B cell activation in murine spleen cells which was comparable to the LPS of P.gingivalis. In contrast, fimbriae from P.gingivalis were not or were weakly effective in murine spleen cells. C3H/HeN mouse macrophages exposed to 10 μg/ml of the 75K protein released high levels of interleukin-1 (IL-1), whereas IL-6 activity in macrophage supernatants was not detected throughout the culture period. These results suggest that the 75K protein and LPS from P.gingivalis are the potent polyclonal B-cell activators and that stimulate IL-1 production from murine peritoneal macrophages, which may therefore play an important role in the inflammatory response in the development of periodontal diseases. |
