Abstract : 「抄録」 アパタイトに唾液蛋白(耳下腺唾液蛋白質)を吸着(pH8.00, μ=0.003, 24hr. 4℃)させた結果, 複数個の吸着蛋白質分屑の存在をDisc電気泳動法により確認した. この吸着耳下腺唾液蛋白分屑(APP)のアミノ酸組成はGlycine, ProlineおよびGlutamic acidを主要構成アミノ酸とし全体の63.8%をしめていた. これは全耳下腺唾液, 獲得被膜(Acquired pellicle)とも組成の異なるPoly-peptidであった. 吸着耳下腺唾液蛋白質(APP)をアパタイトに吸着させたまま, リン酸Na緩衝液(pH8.02, μ : 0.003)において, トリプシンにより部分分解を行なった結果, 吸着ペプチドの主要構成アミノ酸はGlycine, Proline, Glutamic acidおよびSerineであり, 全体の68.3%を占めていた. さらに吸着ペプチドは, ペーパークロマトグラフィー, 高圧電気泳動法によって, 5個の分屑に分離された. 各ペプチドの主要構成アミノ酸は, 酸性および水酸基を有するアミノ酸であるGlutamic acid, Aspartic acidおよびSerineであり, 中性アミノ酸としてGlycineおよびProlineであった. 従って, アパタイトの吸着構造は多元的でありAnionicな反応基の立体配置を強く要求するものと推定される.

Abstract : Attempting to elcidate a steric relation of binding site which might existed on a surface of apatite crystal, human parotid protein-complex were incubated with synthetic hydroxyapatite in phosphate buffer (pH 8.0, μ=0.003) at 4℃ for 24 hours. The adsorped salivary proteins were not, electrophoreticlly, a single fraction, and Glycine, proline and Grutamic acid were the major constituting amino acids which were occupied 63.8 per cent of total 1000 amino acid residues. However, the amino acid composition of this proteins were different one comparing with whole parotid salivary proteins and an acquired pellicle. A peptide chain which were not concerned with binding to the apatite were digested with trypsin (phosphate buffer, pH 8.02, μ=0.003) and resulting peptide conteined Glycine, Proline, Glutamic acid and Serine as the major amino acids which were account for 68.3 per cent of total 1000 amino acid residues. It was worth to note that the adsorped peptide chain were separated into five fragments using the peptid map method and acidic and hydroxy group containing amino acids such as Glutamic acid, Asparatic acid, Serine and Threonine were occupied about 40-60 per cent of total 1000 amino acid residues. Therefore, it was possible to postulate that ther must be more than one adsorping site on the surface of apatite crystal which might mainly consist of cationic charactalized sites.
