Abstract : 「抄録」 卵白リゾチームを吸着(pH7.5, μ=0.004, 2hr, 4℃)させた結果, Lysozyme/Apatite=1/100において85.0%の吸着が明らかになった. この吸着リゾチームをアパタイトに吸着したまま, リン酸ナトリウム緩衝液(pH7.5, μ=0.004)において, trypsin部分分解を行った結果, 吸着ペプチドの主要構成アミノ酸はaspartic acid, glycine, glutamic acidおよびserineであり, 全体の42.5%を占めていた. さらにこのペプチドは, ペーパークロマトグラフィー及び高圧電気泳動法によって, 人耳下腺唾液の吸着分屑同様5分画が存在した. これら吸着部分に極在するペプチドの構成アミノ酸は, 極性アミノ酸であるaspartic acid, glutamic Acid及びSerineが220~420残基/1000残基を有していることは, 卵白リゾチームと人耳下腺唾液のアパタイト吸着分屑とは極めて類似した極性アミノ酸組成を有することが判明した.

Abstract : An apatite, main mineral constituent of teeth, adsorbed an egg white lysozyme in sodium phosphte buffer (pH 7.5, μ=0.004) at 4℃. A portion of the peptides which might interact with the surface of apatite, were obtained with a partial trypsin degradation and five peptides were isolated using paper chromatography (BuOH: AcOH: Water=3:1:1) and high voltage electrophoresis (2000v, 50cm, solvent; pyridine: AcOH: Water=1:10:259). The amino acid composition of these peptides were abundant in residues having anionic group, such as asparatic acid, glutamic acid and serine ranging from 220 to 420 residues out of total 1000 residues of amino acids. The peptides obtained from egg white lysozyme and human parotide saliva possessed an almost identical, as far as, polar amino acids an concerned but dissimilar in neutral amino acids. Therefore, these findings suggest that the steric relation or comformation of anionic group of amino acids are very importance for fullfilment of adsorption between peptides and apatite crystal surface.
