Abstract : 「I 緒言」う蝕とdextranとの関係は, 1965年Carlssonらがsucroseを摂ると歯垢中に多量の細菌菌体外多糖類が生成されることを示したのにはじまり, ついで, 1966年Gibbonsらは, 動物実験でう蝕原性連鎖球菌はsucroseより多量の粘着性の菌体外多糖類を作るが, これに反し非う蝕原性連鎖球菌はほとんど生成しないことを報告している. さらに, 1967年にGibbonsら, Woodら, Leachら, CritchleyらおよびGuggenheimらは, う蝕原性連鎖球菌によってsucroseから合成された菌体外多糖類は, 主としてdextranであり, かつ, それらは歯垢matrixの主要な多糖類であることを示した. う蝕発生の第一段階は, 細菌が歯牙の表面に定着することが必要であるが, このような粘着性の菌体外物質によって菌が歯牙の表面に付着し, 同時に他の細菌をも凝集し歯牙の表面上で増殖する.

Abstract : The dextranase preparation was obtained from broth culture of Chaetomium gracile supplemented with native dextran. Properties of this enzyme preparation, and its use of the polysaccharides produced by Streptococcus mutans and of human dental plaque were studied. The optimum pH of this dextranase was 6. 7. This enzyme was stable within the range of pH 7-11, until 40 C to heat and found to be endo-type dextranase by the results of the increase of reducing sugar, the decrease of viscosity and the thin layer chromatography of dextranase-dextran reaction mixture. This dextranase also showed to α-1,4 glucanohydrolase activity. Moreover, the enzyme showed to α-1,3 glucanohydrolase activity was concomitant in this dextranase preparation. This dextranase preparation inhibited the artificial plaque formation and broke down the polysaccharides produced by Streptococcus mutans, and degradated human dental plaque.
