Abstract : 「緒言」 1928年FreyおよびKrautにより科学的にとりあげられたKallikrein(E C 3. 4. 4. 21)はその後の研究で生体各所で産生され, 血漿中ではその不活性前駆体酵素であるPrekallikreinとして存在することが明らかにされた. そしてこのPrekallikreinは血液凝固第XII因子であるHageman factorやHageman factor fragmentsなどにより活性化されたKallikreinとなり, 血漿中のKininogensから活性PeptideであるKininsを特異的にかつ速やかに遊離させる. このKinins遊離はKallikreinのProtease作用によるものであり, Kallikreinの生理活性は遊離されたKininsの活性として把握することができる. 遊離されたKininsは極微量で生理活性を示し, 熱浮腫, 関節リウマチ, 急性膵炎, ショック等に関与していると考えられているが, これら疾患でのKininsの関与を疑問視する報告も見られる.

Title : A study on Kallikrein-like Esterase Activities in Human Oral Fluids.

Abstract : To investigate the role of kallikrein in periodontal disease, the amount of kallikrein in human whole saliva and gingival fluid taken from patients afflicted with periodontal disease was examined with benzoyl arginine ethylester (BAEe). To obtain the samples for analysis, 36 subjects were selected based on clinical symptoms and roentogenograms and then divided into four groups. Whole salive and gingival fluid were collected from each subject, and the amount of kallikrein were determined for 1.) the whole saliva and 2.) the gingival fluid. The following results were obtained. 1. The hydrolysis rates of BAEe at initial velocities for varying amounts of kallikrein continued to rise uniformly for a period of 30 minutes. 2. The quantitative detection range of human plasma kallikrein was from 0.4 to 3μl, and the kallikrein existed in an inactive form. 3. The quantitative analysis of whole saliva kallikrein was possible from 0.05 to 0.2ml, but in this case the kallikrein existed in an active form. 4. There were high correlations between total kallikrein contents in whole saliva and both clinical symptoms and patients' age. 5. The range of total kallikrein in gingival fluid was from 2.8 to 4.2U/ml. No significant correlation was found between the amount of kallikrein and the degree of periodontal disease or any symptoms.
