Abstract : 「1 緒言」 歯根膜組織は歯と歯槽骨と連結し, 生理的に常時咬合力の加わる強靱な結合組織である. 他の結合組織と比較して, 咬合力の存在下で何故に異なる特徴的な代謝系をもつかを知ることは重要な課題であり, 非生理的状態である歯周疾患の病因論の解明に必須のことと考えられる. そこで著者らは, 細胞の基本的代謝系であり, かつエネルギー産生経路でもあるTCA cycleに関与する有機酸の動的構成は細胞全体の機能からみても特徴的な位置づけを有するものと考えられ, 細胞機能を検索するための1つの重要な糸口となるものと考える. しかし, 歯根膜においてこの関点から追求した検索は殆んど行なわれておらず, 斉藤らによる歯根膜の呼吸量測定において, TCA cycleの中間代謝物質であるsuccinateの添加が呼吸量を大きく増加させるという歯根膜の代謝性の特徴を示唆する重要な現象を確認した報告があるにすぎない. 一方, 口腔内の他の組織である歯髄においてsuccinate添加によるミトコンドリア分画の呼吸量測定, さらに歯肉におけるwhole cellsである組織切片レベル及びミトコンドリア分画について同様の呼吸量測定を行なっている.

Abstract : It is vitally important to explore the metabolic nature of periodontal ligament in order to understand both the physiological and also pathological characteristics of the tissues. Therefore, based on the fact which had reported by S.Saito that the addition of succinate to the bovine incisor periodontal ligament induced extreme increasment of the oxygen consumption about 6-7 times more than that of the endogeneous respiration, the characteristic pathway for the succinate metabolism of bovine periodontal ligament, comparing to the pulp and liver, was studied further by the quantitative analysis of organic acids relating to the TCA cycle and CO2 production derived from succinate-1,4-14C which was added to the short incubation systems as radioactive tracer. The added succinate-1,4-14C was effectively incorporated into the cells of periodontal ligament (78.6%) and oxidatively decarboxylated within the cell which was acounted for 18.0% of added succinate during the 60 min incubation period. Therefore, the rate of CO2 production to the whole organic acids derived from the radioactive succinate was determined as 0.560 which was relatively low value compared to the pulp (0.670) and liver (0.834). However, so far as the individual metabolic activity of those tissue cells based on the amount of substances except the succinate itself, the periodontal ligament did reveal that 44% of the added succinate were converted into the other intermediate which might be utilized for certain other metabolic pathways, while the pulp and liver showed completely differrent ability such as 32.7% and 16.6% respectively. Moreover, the main organic acids synthesized from those tissues during the 60 min incubation were determined as fumarate(F) and malate(M) and the synthetic ratio of these organic acids (F/M) were about 3, whereas the those from the pulp and liver were 2 and 4 respectively. As the results, it is reasonable to elucidate that the periodontal ligament possesses a considerably active and specific potentiality concerning to the utilization of succinate-1,4-14C comparing to the other tissues like the pulp and liver.
