Abstract : 「1. 緒言」 根管充填の時期は, 自覚的他覚的症状が消失し, しかも, 根管の無菌性が獲得された時が望ましいとされている. 根管の無菌性を検査するには細菌検査によるほかはなく, 無菌検査法には培養試験と塗抹標本による検査法とがある. しかしながら塗抹検査法は染色, 鏡検という頻雑な操作を必要とし, 加えて生菌か死菌かの判定が困難とされる. そのため, 一般臨床における根管の無菌性試験には, 根管内容の培養試験がchair timeも少なく, 簡便な方法のため従来から広く用いられてきた. また培養試験は, 根管治療における無菌的処置を習得するには欠くことのできない検査法とされている. 通常, 根管充填が行われる時期には根管内の細菌数は極端に減少し, また細菌自体も薬剤などにより脆弱となっているため, 根管内細菌の状態を確実に反映させるには, 優れた培地と細菌の増殖に最も適した温度を常に保つような培養器が必要となってくる.

Title : A Comparative Study of Various Incubators for Culturing Microorganisms from Root Canals.

Abstract : [Abstract] : In determining the sterility of pulpless teeth prior to root canal obturation, the culturing test has been widely applied as a practical method. The results obtained from the test, however, are largely dependent upon such factors as bacterial contamination in the process of cultivation, the formula of the applied culture medium, the efficiency of the used incubators, etc. With a view to evaluating the efficiency of incubators, a comparative examination was made in this study by picking up four kinds of incubators A, B, C, and D, each being commercially available under a different brand. The experiments were carried out to investigate the changes of inner temperature of each incubator when it was kept under room temperature (22℃) and lower temperature (4℃), as well as to examine the microorganism growth under different room temperatures by using Staphylococcus aureus (209P), Streptococcus salivarius, Candida albicans and Veillonella as the reagents. The observed results were summarized as follows : 1) A better growth of Staphy. aureus was observed in Incubators A and C than in Incubators B and D, While no remarkable difference was observed as to Strept. salivarius growth in the four incubators. The growth of Candida albicans in Incubators B and D showed an inferior result as compared with the case of Incubators A and C. 2) The results of thermal cycling test revealed that Staphy. aureus in Incubator A grew better than in the other three, whereas no difference was observed as to strept. salivarius growth. Candida albicans showed the similar inferior growth in the four incubators alike, while as to Veillonella, scarcely any growth was observed in Incubator B, even when a high density reagent was applied, and a better growth result was obtained by Incubator D than by Incubators A and C. 3) From the viewpoint of the changes of inner temperature, it was maintained almost at 37℃ regardless of the outside thermal changes in the case of Incubators A and C, but the inner temperature of Incubator B changed respectively from 35℃ to 42℃ at room temperature and from 35℃ to 45℃ at a low temperature. In the case of Incubator D, the inner temperature changed from 32℃ to 33℃ at room temperature, but it remained at the constant low level of 21℃ at a lower outside temperature. 4) The foregoing observations suggest that a type of equipment like Incubator A or C in which the inner temperature can be maintained at a stabilized level is better fitted for the purpose of culturing microorganisms of root canals.
