Abstract : 「抄録」 軟口蓋と硬口蓋の組織構造の差異を明らかにするため, 血管の分布と肥満細胞を中心にWistar系ラットを用いて検索し, さらにhistamine liberatorのCompound48/80投与による軟口蓋及び硬口蓋のhistamine型血管透過性亢進に対する反応性の差異について検討を加えた. 軟口蓋と硬口蓋においては血管の走行と肥満細胞の数および性状に著しい相違が見られた. すなわち軟口蓋は静脈性血管網に富んでおり, 肥満細胞の数が多く, これに反して硬口蓋は静脈網の発達が弱く, 肥満細胞の数も極めて少なかった. Compound48/80投与により軟口蓋は著明に腫脹し, 生体染色剤Pontamine sky blueの強い青染を示すが一方, 硬口蓋での青染は少なかった. 組織化学的検索でCompound48/80投与により軟口蓋では静脈系, 特に細静脈に著明な血管透過性亢進が見られ, 血管周囲の肥満細胞はすべて脱顆粒現象を生じたが, 硬口蓋ではほとんど脱顆粒現象は認められなかった. 口蓋粘膜についてAlcian blue-Safranine重染色により肥満細胞の性状を検索すると, 軟口蓋にはSafranine陽性の成熟型の肥満細胞が多数存在し, 硬口蓋の肥満細胞はAlcian blue陽性の未熟型で, その数も少なかった. 更に電顕的にもこれらを裏付ける所見が得られた. 螢光測光法による検索では, 軟口蓋にみられる成熟型の肥満細胞はhistamine, serotoninを多量に含有し, Compound48/80投与によって両者の著しい放出が見られ, 反応性が高く, それに反し, 硬口蓋に少数ながら見られる未熟型の肥満細胞は, histamine, serotoninの含有量が少なく, またCompound48/80に対する反応性も低いものであった. 以上の結果から, ラットにおいては硬口蓋に比較して, 軟口蓋にはよく発達した静脈網を認め, これに近接して反応性の高い成熟型の肥満細胞が多数常在しており, 炎症性の刺激に対して直ちにhistamine型血管透過性亢進を惹起せしめる可能性が示唆されると同時に, ラットの軟口蓋は生体の防御的な面において重要な意義を有するものと考えられる.

Title : A Study of the Distribution and Development of Mast Cells and Blood Vessels in the Soft and Hard Palate Tissues and Their Reactivity to Compound 48/80.

Abstract : To elucidate the differences in the structure between the soft and hard palate tissues, the distribution and development of the blood vessels and mast cells of the palates were investigated using Wistar strain rats. Differences of the histamineinduced vascular permeability between the soft and hard palate tissues after the administration of histamine liberator compound 48/80 were also investigated. Number and configurations of blood vessels and mast cells in soft and hard palate tissues were markedly different. The venular network was developed to a higher degree and the number of mast cells in the vicinity of the venule was larger in the soft palate tissues than those in the hard palate tissues. When the compound 48/80 was administered, the soft palate tissues swelled significantly and were stained blue with the pontamine sky blue. Histochemical examination showed a marked increase of the permeability of the venule in the soft palate tissues and a degranulation of the mast cells around the venule after the administration of compound 48/80. Essentially no such changes were observed in the hard palate tissues. When doublestained with alcian blue and safranine, there were many safranine-positive mature mast cells in the soft palate tissues, while in the hard palate tissues, there were only alcian blue-positive immature mast cells. These results were also supported by electronmicroscopy. Quantitative microfluorimetry showed that the mature mast cells in the soft palate tissues contain large quantities of histamine and serotonin. In the compound 48/80 administered rats, the content of histamine and serotonin in the mature mast cells in the soft palate tissues was reduced significantly, proving that these mast cells have a high response to compound 48/80. In contrast, the immature mast cells in the hard palate tissues contain small quantities of histamine and serotonin. These immature mast cells have a low response to compound 48/80. Thus, our experiments proved that the venular network develops better in the soft palate tissues than that in the hard palate tissues. These findings suggest that when an inflammatory stimulus is given, the vascular permeability is enhanced more quickly in soft palate tissues than that in hard palate tissues.
