Abstract : 「緒言」 う蝕は, 歯周疾患とならび口腔内の二大疾患のうちの一つである. 特にう蝕は, 小児期から高い有病率を示し, 昭和50年度歯科疾患実態調査報告によれば, 乳歯のう蝕有病者率は1才で11.4%, 2才で51.7%, 3才で84.2%, 永久歯のう蝕有病者率は, 6才で36.0%, 7才で66.3%, 10才ですでに90%を超え, 12才以後は, 40才代前半までにそのほとんどの年齢で95%を超えている. しかし, ごく最近では小児のう蝕罹患状況は, 徐々に改善されつつある. このことは, う蝕予防に関して基礎的研究, 臨床面あるいは行政面における活動が, 積極的に推進されている結果であろう. う蝕予防処置あるいは歯科保健指導に際して行なわれるう蝕活動性試験は, 口腔環境を改善するための動機づけとして極めて有効な一手段である. 従来から, 種々のう蝕活動性試験が考案されており, 例えば, 唾液中の乳酸桿菌数の測定試験, スナイダーテスト, 唾液緩衝能の測定試験, プラークのpHの測定試験, エナメル質溶解試験, 唾液成分の乳酸桿菌に対する抗菌試験などの多くの試験法が研究されてきた.

Abstract : M S agar is generally used as a means of isolating streptococci from the mouth. It shoud be recognized, however, that it is difficult to isolate Str.mutans among the oral microorganisms. Recently, the M S medium modified by addition of a few antibiotics, growth inhibitors and excessive sucrose has been reported. It is known that these media inhibit not only the growth of Str.mutans, but allow the multiplication of other bacteria. Therefore, we have attempted to develop a highly selective medium. MATERIALS, METHODS AND DISCUSSION The medium contains an indicator dye, bromocresol purple which changes from purple to yellow in the range of pH 6.8 to 5.2. The optimum concentration of growth inhibitors in this selective medium, phenethyl alcohol and sodium azide, were 0.1% and 0.0075%, respectively. Mannitol was utilizied as a fermentable carbohydrate. When using this newly developed medium to isolate Str.mutans from human plaque, large yellow colonies were observed on the agar plate. These colonies proved to be Str.mutans biologically and morphologically. Presently, we are interested in the application of this medium named "K K Y" for caries activity testing.
