Abstract : 「緒言」 歯肉溝における滲出液の存在については, 1958年Brillが犬の静脈内に蛍光物質を投与すると上皮下結合組織を通じて, 歯肉溝へ移行する事実を報告して以来, 歯肉溝滲出液に関する生理学的および生化学的研究が多く報告されている. すなわち, 歯肉溝滲出液量については, 生理的にも異なるがホルモン性変化, 酵素処理ブラッシング, 歯肉マッサージ, および局所炎症状態により変化することが報告されている. 一方, 滲出液に含有される物質に関しては, アミノ酸および血清蛋白が血中より歯肉溝へ滲出していると報告している. 歯肉溝滲出液に含有される遊離アミノ酸は主にGlutamicacid, Alanine, Serine, Cystineといわれている. また滲出液中の蛋白質は, Albumin, α1-globulin, α2-globulin, β1-globulin, γM-globulin, γG-globulin, γA-globulin, およびfibrinogen等が検出されている.

Abstract : Abstract : Many investigators have already established the fact that dental plaque is a main cause for the initiation of periodontal disease. This bacterial stimulation causes the gingiva to exhibit continuous serial changes, which are referred to as inflammatory responses. Therefore, the gingival fluid, through crevicular sulcus, may contain varied levels of inflammatory products, such as the degradative products of tissue and plasma proteins. In this study, macromolecular proteinous componets in the gingival fluid were analyzed by acrylamide gel electrophoresis. This method was modified by the author in order to improve electrophoretic resolution for a trace amount of specimen. To understand the chemical nature of the degradative products, amino acids analysis was performed on 10% TCA insoluble, soluble, and free amino acids using an automatic amino acids analyzer. The data revealed that proteinous residues and supernatant peptides of 10% TCA solution showed characteristic changes in their amino acids composition corresponding to the gingival status. The relative amoumt of serine for free amino acids was decreased remarkably. This decrease corresponded to an advancement of inflammaton which indicated the acceleration of hydrolytic cleavages of proteoglycans, the main noncollagenous porteins in the gingival connective tissues. The author therefore concluded that the analysis of gingival fluid from clinically healthy gingiva, gingivitis, and marginal periodontitis may provide an objective and important information for the diagnosis of periodontal diseases.
