Abstract : 「緒言」 歯周疾患は, 口腔内細菌と宿主細胞との間で起こる複雑なHost-parasie relationであり, その結果生ずるコラーゲン及びglycoconjugatesを主要代謝産物とする結合組織線維芽細胞の代謝変動(metabolic alteration)とも言える. この様な病変形成の過程において, 細菌由来の外来物質が炎症反応のTriggerあるいはmediatorとしての役割を演ずるだけでなく, 不都合なる免疫応答も充分に考慮せねばならないことは周知の事実である. さらに, これら生体機序を理解する基本として, 細菌由来物質と標的細胞の代謝的変動の把握が要求されるが, その知識はまだ未開発の段階にある. EIIison(1970)は歯垢中には多数の細菌が存在しているが, 細菌が直接歯周組織内に侵入し組織を破壊するという病態像はないことを報告している. また, Socransky(1970)はこれらのことから歯垢中の細菌が産生する浸潤性かつ細胞毒性を有する物質が, 歯周組織破壊に関与する可能性を示唆している.

Title : Study of Metabolic Alteration of Bovine Periodontal Ligaments by Endotoxin Effect of E.coli Endotoxin on Biosynthesis of Macromolecules and Glycosaminoglycans

Abstract : Abstract : The metabolic alteration caused by the endotoxin, Lipopolysaccharides E.coli 0127 : B8 (LPS), upon the synthesis of glycosaminoglycans (GAGs) has been investigated by quantitative analysis of radioactive macromolecules. The radioactive macromolecules were converted from succinate-1,4-14C according to the short term incubation method and by using freshly extracted bovine periodontal ligaments. The non-dialyzable fractions of the incubation medium were chromatographed by Sepharose CL-6B and the radioactivity of exclusive macromolecules showed a remarkable increase, whereas, no difference was observed for inexclusive fractions. Compared to that of the control value without LPS, the increase of the macromolecules occurred according to the LPS dosage in terms of either the absolute or relative values of the radioactivity. This fact indicates that a certain pathway of proteoglycans synthesis is altered by the endotoxin ; however, it is interesting to note that no regulatory alterations are observed for the synthesis of glycoproteins. Thus, the electrophoresis of the GAGs was performed after pronase digestion of the proteoglycans according to the routine method. The results obtained in this experiment demonstrate an increase of 21% in the total GAGs due to the LPS stimulation when compared to the control. The increase can mainly be explained by the acceleration of the hyaluronic acid synthesis. The control value of the hyaluronic acid was (70.6+-4.3)×10 cpm/g tissue. there was no inhibitory reaction observed throughout the synthesis of various GAGs ; however, with the addition of LPS the value of the hyaluronic acid increased to (103.8+-11.1)×10 cpm/g tissue. This data indicate the fact that the LPS induced the extraordinary enhancement of the specific enzymatic activity relating to the glyconeogenesis.
