Abstract : 「緒言」 歯周疾患は歯垢付着に伴う歯周結合組織の病変であり, Pageらはこれらの病変を臨床病理組織学的に初期initial stage, 早期early stage, 確立期established stage, 極期advanced stageの四期に区別している. 即ち初期initial stageにおいては歯垢付着に伴う付着上皮下の急性滲出性炎症を特徴とし歯垢成分による直接の組織為害作用と多形核白血球を主体とする食細胞系の非特異的炎症反応が生ずる. early stage早期においてはより顕著な細胞浸潤と結合織成分の崩壊を示すが, 付着上皮下結合組織における細胞浸潤はリンパ球(T-cell, B-cell)を主体とする免疫担当細胞の浸潤であり, 以後プラズマ細胞の浸潤を主体とした確立期established stageポケット形成, 歯槽骨の吸収を呈する極期advanced stageに進展して行く. 特に早期early stageにおけるリンパ球を主体とする浸潤細胞は一連の免疫学的な作用を発揮し, 細胞性免疫が成立しているものと考えられる.

Abstract : Abstract : Much research has been performed in order to understand the pathogenic mechanisms of chronic inflammation in relation to cellular immunity. For the purpose of simplifying the complexity of cell to cell interaction in a region of inflammation, model experiments were designed to study the interaction between porcine peripheral blood lymphocytes and L929 fibroblasts by the utilization of the cell culture system. In this experiment, porcine peripheral blood lymphocytes were stimulated by phytohemagglutinin, a non-specific mitogen. Limphokines, especially those produced during 24 to 48 hours, were added to mouse L929-fibroblasts culture medium. Both activities of lysosomal enzymes released to the medium and sustained within the cells were estimated by assay in the typical lysosomal enzyme such as acid phosphatase, β-glucuronidase, and cathepsin B1. Conclusive data showed that the released acid phosphatse activity increased 3.5 times and the intracellular activity intensified 4 times when compared with the controls. Also, the released β-glucuronidase activity showed an increase of 2.5 times and the intracellular activity increased 3 to 4 times. These results suggest that the lymphokines, which are produced by PHA-P stimulation during a period of 24 to 48 hours, have a biological effect of promoting L929-fibroblasts in order to release the lysosomal enzymes as well as accelerating their biosynthesis.
