Abstract : 「緒言」 口腔粘膜の実験モデルによる基礎的研究は1954年Salleyの報告により飛躍的な進歩をとげた. Salleyはハムスター頬嚢で3種のcarcinogenic hydrocarbonについての発癌性を比較し, 9,10-dimethyl-1,2-benzanthracene(DMBA)が最も発癌効果の高いことを報告した. 以来, Salleyによって記載された方法に基づき, Morris(1961)は実験動物の性と年齢について検索し, 若いハムスターが発癌性が高く, 性差はみられないことを報告した. また, Dachiら(1962)(1967)は発癌剤DMBAの濃度と溶媒について検討し, 癌発生が発癌剤の濃度に依存し有意差がみられることと, 癌発生からの生存期間が使用した溶媒によって異なることを報告した. その他, 癌化に対するVitaminの影響についてRowe(1959)はVitamin Aの欠乏を, Salleyら(1962)はVitamin Bを欠乏させた場合を検索し, いずれも発癌促進効果がみられることを証明している.

Title : Pathological Study of Experimental Buccal Mucosal Carcinoma in Hamsters

Abstract : Abstract : Carcinma of the buccal was occurred in hamsters by abrading them with a dental cleanser and applying a chemical carcinogenic agent, 9,10-demethyl-1,2-benzanthracene (DMBA). The development of this carcinoma was studied from the standpoint of pathologic anatomy. 1. This process was divided into the following 6 stages according to their macroscopic and histological findings ; i) rednesss-swelling stage, ii) ulceration stage, iii) thickening of the mucous membrane stage, iv) small tumor formation stage, v) cancer stage, and vi) advanced cancer stage. 2. In the first cancerous period in this experimental cancer, the ulceration was always observed. This ulceration stage plays an important role in the onset of the concer and in determining the tissue type of the cancer. 3. Cancer developed in this experiment presented as two distinct types ; a well differentiated type and a poorly differentiated type of sbuamous cercinoma. Well differentiated types of squamous cell carcinoma arose from lesions resembling human leukoplakia. Poorly differentiated types of squamous cell carcinoma primarily developed from fibroid polyps arising from an ulcerative region, and represented downward proliferation and infiltration. 4. This experimental cancer developed multiply and descontinuously, and occurred centmultiply. 5. Dark cells with high electron donsity and light cells with low electron density were observed electron-microscopically. The dark cells contained abundant ribosome. In addition, the dark cells contained abundant or few tonofilaments. 6. Abnormally keratinized cells and single keratinized cells were frequently observed in the well defferentiated type of squamous cell carcinoma. These phenomena were considered important in the diagnosis of the onset of the cancer. 7. Metastases to the submandibular lymph node were noted in 2 of 5 hamsters, which were followed for a long time (23 weeks). Histological findings of the metastatic lesions were showed to be poorly differentiated carcinoma. 8. Fibroid polyps were characteristically noted in the development of the experimental cancer. Hemangioma and carciosarcoma also developed. These findings were suggested proliferative effects of DMBA on the fibroblasts and endothelial cells.
