Abstract : Abstract : During 12th of gestation to 8 weeks after birth of the rats, the differentiation and the distribution of mast cells in the oral region were examined by the light microscope and the electron microscope. The following results were obtained. 1. Mast cells initially appear in the loose connective tissue under the skin of the back and in the caudal area in 15-day-old fetuses. 2. At 1 day after birth, rapid increase of mast cells was observed in the oral region, perineum, testes and perianal region. 3. The distribution of mast cells in the oral region was extremely high in the tongue, submandibular glands and palate. Mast cells rarely appeared in the gingiva, mandibular marrow and periodontal ligaments, and were never observed in the dental pulp. 4. The mast cells distributed in the oral region generally increased until 3 to 4 weeks after birth. Then they tended to decrease slightly, and thereafter there was no variation. 5. The cytoplasm of the mast cells which first appeared in the 15-day-old fetuses was small in size and was contained a few granules. Thereafter, the cytoplasm increased in size and rapidly grew to maturity by 1 to 2 weeks after birth, also showing an increase in the number of intracellular granules. 6. AB-S stain revealed that all mast cells in their first stage of appearance were type I MC. As the day went, the number of mast cells containing safranin positive granules increased, and reciprocally the number containing alcian blue positive granules decreased. At 2 weeks after birth, most mast cells in rats were type V MC. 7. AB-S stain was not uniform in the palate even in rats 2 weeks after birth. Only type I, II and III MC were present in the hard palate, and type III and IV MC were observed in the dental alveoli, while most mast cells in the soft palate were type V MC. 8. Using fluorescent histochemical observations, the mast cells in 15-day-old fetuses were already found heparin and serotonin. Histamine, in rats, was observed one day after birth.
