Abstract : 「緒言」 歯科領域における咬合の重要性は非常に大きな問題である. そこで, 咬合力が歯周組織に如何なる代謝的影響を与えているかを把握することは, 咬合状態の良否を決定する一つの大きな要因を識るものと考えられる. 秋吉, 佐伯, 井上らは, 歯が対合歯と正常な咬合関係を有している時にのみ, 歯周組織は一定の機能構造を形成し, ウ蝕やその他の原因により対合歯との正常な咬合関係を喪失すると, 歯根膜の狭窄, 歯根膜線維の減少と走向の乱れ, 根間中隔頂部を中心とする骨添加および根尖部を中心とするセメント質添加等の廃用性萎縮と総称できる変化を報告している. さらにPihlstromら, 吉野も同様な報告をしているが, 佐伯は, ラットの大臼歯の歯周組織における実験的廃用性萎縮の発生過程およびその修復過程について, 経時的な組織学的検索を行ない, 対合歯との咬合関係を喪失後48時間で根分岐部に既にsharpey線維の減少が発現しており, その部の歯根膜は疎性になっていることを報告している.

Abstract : Abstract : In order to understand the direct relationship of occlusion to periodontal diseases, investigations involving the research of metabolism of periodontal ligament under hypo-functional occlusion have proven to be most important. The basis of this investigation was founded upon the research conducted by S.Saito which demonstrated the involvement of oxygen consumption by the means of the addition of succinate into the bovine incisor's periodontal ligament. These results showed an increase of oxygen consumption of about six to seven times. Periodontal ligament dissected from hypo-functional occlusion of the rat was analyzed by the short term incubation system by using succinate-1,4-14C as a radioactive tracer. This analysis was performed in order to find the metabolic alteration of organic acids, amino acids and macromolecules due to occlusive forces. The results obtained were as follows : 1) By incubation with succinate-1,4-14C, uptake of succinate was 38.5% at control and 43.4% at hypo-functional occlusion occurring for 12 hours (12 hours period) after removal of occlusive surface. 2) The main organic acids synthesized from periodontal ligament were fumarate and malate. Malate increased about 1.5 times after the 12 hours period. 3) The main amino acids derived from the TCA cycle were Asp., Glu., Gly., Ala., Ile., Leu. and Phe. Acidic amino acids increased about 3.5 times after the 12 hours period, but neutral amino acids decreased about 4/5. 4) Macromolecules decreased about 2/3 after the 12 hours period and the pattern of Sepharose CL-6B gel chromatography changed after the 12 hours period compared with the control.
