Abstract : 「緒言」 ラット顎下腺中には, testosterone(T)を活性型androgenであるdihydrotestoterone(DHT)に変換する5α-reductaseが存在すること, およびDHTの蓄積があることなどから, androgenの標的組織であるといわれている. さらに, 顎下腺がandrogenの標的組織であるためには, 顎下腺内にandrogenにたいするreceptorが存在することが重要な要素である. Wilsonらは, マウスを用いた実験で, 顎下腺にDHTと特異的に結合するタンパクが存在すると報告している. 一方androgen receptor assayは, 主に前立腺について検索が進められてきた. しかし, このreceptor assayにおいて問題となるのはTeBG(Testosterone-Estradiol Binding Globulin)の存在である.

Abstract : Abstract : The binding of methyltrienolone (R1881) in rat submandibular gland (SMG) cytosol were studied by a charcoal assay. R1881 binding complex were stable at 0℃ for 20hr, showing neither dissociation nor metabolism. Binding of dihydrotestosterone (DHT), estradiol (E2), testosterone (T) and moxestrol (R2858) were small, while R1881 bound significantly to the cytosol of rat SMG. This R1881 binding were competed by unlabeled E2 and DHT, and most effectively by progesterone (P4). The effect of triamcinolone acetonide, the inhibitor of R1881 binding to progestin receptor, was not shown in cytosol of the SMG. The apparent Kd of the binding to R1881 was determined by Scatchard analysis. Kd were 3.51nM for male and 4.82nM for female. NBS (number of binding site) of R1881 were 262 fmol/mg protein for male and 1060 fmol/mg protein for female respectively. From these studies, it was shown that the R1881 binding substance were containd in SMG cytosol of both male and female rats, and this complex were not contaminated by progestin receptor. Further this substance had high affinity for R1881, and capacity of the substance in female rat were higher than male.
