Abstract : 「はじめに」 近年, 生理活性物質に関する研究の進歩は目覚しいものがあり, 極く微量で効果を示す物質が数多く発見されている. これら活性物質の微量定量法や精製法の開発, 改良といった化学的領域における発展は, 生理学, 生化学, 薬理学あるいは内分泌学といった広い分野でその役割を果たしてきた. 生理活性物質に関する研究の対象は, peptidが主であり, 内分泌化学の発展に伴い, 生体制御物質として, その役割が明らかにされてきた. Vasopressin, oxytocin, ACTHのような下垂体hormoneあるいはinsulinなどがその良い例である. 現在, 臓器分泌系hormoneの大部分は構造レベルまでも解明され, さらには, morphine様peptideと呼ばれる全く新しい生理活性peptideが注目されるに至った. しかも, これら生理活性peptideは治療や診断に用いられるものが多く, 臨床上大いに貢献している.

Title : Relationship Between the Effect of Peptide Extracted from the Skeletal Muscles of Fur Seals on Peripheral Circulation and the Physiologically Active Components

Abstract : Abstract : In the previous study, it has been confirmed that physiologically active peptide extracted from the skeletal muscles of fur seals has beneficial effect on inflamed periodontal tissues. This study was designed to investigate the mechanisms of the effect of the peptide in canine peripheral circulation and in the microcirculation of hamster cheek pouch. Further, the vasoactive fraction of the peptide was purified for examination between the structure and the activity. The blood flow in gingiva, carotid artery and femoral artery were increased by the peptide in a dose-dependent manner. These increases were prevented in part by atropine, diphenhydramine or by propranolol. This indicates the peptide has muscarinic, histaminic or β2 adrenergic action on circulatory dynamics. Dilation of microvesseles in hamster cheek pouch was induced by the peptide. This effect in capacitance vessel was more potent than in resistance vessel. The circulatory effect of the peptide stated above was more potent than that of carnosine. About 6 components of vasoactive peptide were found in the peptide extracted from the skeletal muscles of fur seals. Various chromatography revealed that molecular weight of physiological active component of the peptide is less than 500, and that the eluted fraction of this component is the same as that of neutral peptides.
